Horseradish peroxidase, abbreviated as HRP, is a single-chain polypeptide containing four disulfide bridges. It is a glycoprotein consisting of 18% carbohydrates. The composition of these carbohydrates includes galactose, arabinose, xylose, fucose, mannose, mannosamine, and galactosamine, depending on the specificity of the isoenzyme. HRP consists of several isoenzymes with a molecular weight of 44,000 Da, structured by polypeptide chains (33,890 Da), heme plus Ca2+ (700 Da), and carbohydrates (~ 9,400 Da). HRP has an isoelectric point of pH 3–9, and the optimum pH for enzyme catalysis varies slightly depending on the hydrogen donor but is mostly around pH 5.
HRP is currently the most widely used labeling enzyme for antibodies and proteins, which can be used in both localization detection and quantitative determination. It has been adopted for various biochemical applications, such as western blotting, ELISA, and immunohistochemistry, and it is applied to amplify weak signaling and enhance the detection of target molecules, such as proteins. HRP is commonly used to determine the amount of glucose and peroxide in solutions and has also been used for rapid susceptibility testing in Aspergillus fumigatus.
